Tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor, making it a dual agonist rather than a selective agent. Engagement of the GLP-1 receptor is linked to glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. The relative contribution of the GIP arm remains an active research question; proposed roles include improved insulin sensitivity and altered adipose tissue handling. Receptor occupancy studies suggest the molecule interacts with both targets at circulating concentrations achieved during therapy.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.

Clinical research programs have evaluated tirzepatide in adults with type 2 diabetes and in adults with obesity or excess weight. Trials generally reported reductions in glycated hemoglobin and body weight across treatment periods of several months. Since these studies enrolled defined populations under controlled conditions, the findings describe group averages rather than individual outcomes. Open questions include the durability of effects after treatment stops, variation among subgroups, and the long-term consequences of sustained dual receptor stimulation. Published trial summaries should be consulted for exact measurements rather than secondary accounts.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its backbone derives from the native glucose-dependent insulinotropic polypeptide sequence, altered at several positions to resist enzymatic cleavage. A fatty diacid group attached through a linker extends plasma residence time by promoting reversible binding to serum albumin. The molecule carries a net negative charge near physiological pH and has a reported molecular weight close to 4813 daltons. These features separate it from shorter incretin analogs and account for its prolonged dosing interval.
Pharmacologically, tirzepatide activates two distinct G protein-coupled receptors: the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Binding at each target triggers cyclic AMP accumulation and downstream signaling in pancreatic beta cells, adipose tissue and the central nervous system. Because the two pathways overlap only partially, the combined effect on insulin secretion, glucagon suppression and appetite signaling differs from that of selective single-receptor compounds. Affinity is not equal across the two targets, and the clinical meaning of that imbalance remains an area of active study.
Recommended storage for reference material is a freezer at approximately -20 degrees Celsius, protected from light and moisture. Commercial injectable presentations are stored refrigerated between 2 and 8 degrees Celsius and must not be frozen. Product labelling generally permits a limited period at controlled room temperature once dispensed, with the exact window depending on the presentation. Repeated temperature cycling is avoided because it can promote aggregation or deamidation of the peptide chain.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
=== Spintronics === Large-area graphene created by chemical vapor deposition (CVD) and layered on a SiO2 substrate, can preserve electron spin over an extended period and communicate it. Spintronics varies electron spin rather than current flow. The spin signal is preserved in graphene channels that are up to 16 micrometers long over a nanosecond. Pure spin transport and precession extended over 16 μm channel lengths with a spin lifetime of 1.2 ns and a spin diffusion length of ≈6 μm at room temperature. Spintronics is used in disk drives for data storage and in magnetic random-access memory. Electronic spin is generally short-lived and fragile, but the spin-based information in current devices needs to travel only a few nanometers. However, in processors, the information must cross several tens of micrometers with aligned spins. Graphene is the only known candidate for such behavior.

== Development == Cutaneous structures arise from the epidermis and include a variety of features such as hair, feathers, claws and nails. During embryogenesis, the epidermis splits into two layers: the periderm (which is lost) and the basal layer. The basal layer is a stem cell layer and through asymmetrical divisions, becomes the source of skin cells throughout life. It is maintained as a stem cell layer through an autocrine signal, TGF alpha, and through paracrine signaling from FGF7 (keratinocyte growth factor) produced by the dermis below the basal cells. In mice, over-expression of these factors leads to an overproduction of granular cells and thick skin. Hair and feathers are formed in a regular pattern and it is believed to be the result of a reaction-diffusion system. This reaction-diffusion system combines an activator, Sonic hedgehog, with an inhibitor, BMP4 or BMP2, to form clusters of cells in a regular pattern. Sonic hedgehog-expressing epidermal cells induce the condensation of cells in the mesoderm. The clusters of mesodermal cells signal back to the epidermis to form the appropriate structure for that position. BMP signals from the epidermis inhibit the formation of placodes in nearby ectoderm. It is believed that the mesoderm defines the pattern. The epidermis instructs the mesodermal cells to condense and then the mesoderm instructs the epidermis of what structure to make through a series of reciprocal inductions.
Myostatin inhibitors are a class of drugs that work by blocking the effect of myostatin, which inhibits muscle growth. In animal models and limited human studies, myostatin inhibitors have increased muscle size. They are being developed to treat obesity, sarcopenia, muscular dystrophy, and other illnesses.

Paracetamol, or acetaminophen, is an analgesic and antipyretic agent used to treat fever and mild to moderate pain. It is a widely available over-the-counter drug sold generically or under various brand names, including Tylenol and Panadol. Paracetamol relieves pain in acute mild migraine and episodic tension headaches. At a standard dose, paracetamol slightly reduces fever, though it is inferior to ibuprofen in that respect: the benefits of its use for fever are unclear, particularly in the context of fever of viral origins. Paracetamol is effective for pain after wisdom tooth extraction, but it is less effective than ibuprofen. The combination of paracetamol and ibuprofen provides greater analgesic efficacy than either drug alone. The pain relief paracetamol provides in osteoarthritis is small and clinically insignificant. Evidence supporting its use in low back pain, cancer pain, and neuropathic pain is insufficient. Paracetamol is the first-line treatment for pain and fever in pregnancy; no causal association with neurodevelopmental disorders has been established, while untreated pain and fever can harm the mother and fetus. At a recommended maximum daily dose for an adult of three to four grams, paracetamol is safe and effective in the short term when used as directed, with uncommon adverse effects similar to those of ibuprofen. It is often used in patients who cannot tolerate nonsteroidal anti-inflammatory drugs (NSAIDs) such as ibuprofen.
== Occurrence and nomenclature == The term nucleic acid is the overall name for DNA and RNA, members of a family of biopolymers, and is a type of polynucleotide. Nucleic acids were named for their initial discovery within the nucleus, and for the presence of phosphate groups (related to phosphoric acid). Although first discovered within the nucleus of eukaryotic cells, nucleic acids are now known to be found in all life forms including within bacteria, archaea, mitochondria, chloroplasts, and viruses (There is debate as to whether viruses are living or non-living). All living cells contain both DNA and RNA (except some cells such as mature red blood cells), while viruses contain either DNA or RNA, but usually not both. The basic component of biological nucleic acids is the nucleotide, each of which contains a pentose sugar (ribose or deoxyribose), a phosphate group, and a nucleobase. Nucleic acids are also generated within the laboratory, through the use of enzymes (DNA and RNA polymerases) and by solid-phase chemical synthesis.
During Alice in Chains' hiatus, reports of Staley's addiction began to gain widespread circulation in fan and media communities, in part due to changes in his physical condition brought on by prolonged heroin abuse. Alice in Chains regrouped to record Alice in Chains, sometimes referred to as "Tripod" due to the cover featuring a three-legged dog, which was released in November 1995. The album debuted at the top of the U.S. charts, and has since been awarded—along with Facelift and Jar of Flies—double platinum status. With the exceptions of "Grind", "Heaven Beside You", and "Over Now", the lyrics were all written by Staley, making this album his greatest lyrical contribution to the band's catalog. To accompany the album, the band released a home video, The Nona Tapes, but the band lapsed again, failing to complete tours planned in support of the album. When asked about the frustration of not touring to support the record, Cantrell provided some insight into how Staley's addictions led to tensions within the band: "Very frustrating, but we stuck it out. We rode the good times together, and we stuck together through the hard times. We never stabbed each other in the back and spilled our guts and [did] that kind of bullshit that you see happen a lot." "Drugs worked for me for years", Staley told Rolling Stone in February 1996, "and now they're turning against me, now I'm walking through hell and this sucks. I didn't want my fans to think that heroin was cool. But then I've had fans come up to me and give me the thumbs up, telling me they're high.
== Nutrition == A raw shallot contains 80% water, 17% carbohydrates, 2.5% protein and a negligible amount of fat (table). In a reference amount of 100 grams (3.5 oz), raw shallot supplies 72 calories and is a rich source of vitamin B6 (20% of the Daily Value, DV), while providing moderate amounts of manganese (13% DV) and potassium (11% DV). No other micronutrients occur in significant amounts (below 10% DV, table).
== Symptoms and signs == Abdominal obesity, often assessed using the waist-to-hip ratio, is associated with hyperinsulinemia. Direct measurement of serum insulin levels can identify elevated insulin, which may precede the onset of impaired glucose tolerance or hyperglycemia by 10 to 20 years. Consequently, normal fasting blood glucose levels alone do not rule out early-stage hyperinsulinemia. Some patients may experience a variety of symptoms when hypoglycemia is present, including:
