Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Research-grade material circulates through suppliers that differ widely in documentation and testing practice, so a certificate of analysis is a starting point rather than proof of quality. Independent verification typically repeats chromatographic purity and mass confirmation on the received lot, and compares results against a retained reference standard. Regulatory status varies by jurisdiction, and a substance cleared as a medicine is not interchangeable with a research chemical of the same name. Open questions include how closely non-pharmaceutical lots match approved material in impurity profile and in aggregate content.
Solid tirzepatide is handled as a lyophilised, hygroscopic peptide powder that should be kept desiccated, protected from light, and stored frozen, typically at or below minus twenty degrees Celsius for long-term retention. Material left at ambient temperature for extended periods can take up moisture, which promotes aggregation and deamidation. Commercial liquid presentations are kept refrigerated between two and eight degrees Celsius and are not frozen. Reconstituted laboratory solutions are generally held cold and used within a short window because hydrolysis and oxidation continue slowly in solution.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography for the main peak and with mass spectrometry for the observed molecular mass. Peptide mapping after enzymatic digestion confirms the primary sequence, while amino acid analysis provides a quantitative composition check. Size-exclusion chromatography and ion-exchange chromatography are used to look for aggregates and charge variants. Water content, residual solvents, and counter-ion content are measured separately, since a lyophilised powder is often reported on an as-is basis unless a correction is applied.
| Property | Value | Notes |
|---|---|---|
| Molar mass | approximately 4813 Da | calculated from the 39-residue sequence |
| Appearance | white to off-white powder | typical of lyophilised peptide material |
| Solubility class | freely soluble in water | measured value depends on salt form and pH |
| Plasma protein binding | >99 percent | linked to the fatty diacid side chain |
| Class | dual GIP and GLP-1 receptor agonist | receptor activity varies with the assay used |
The GIP receptor is expressed in pancreatic islets, adipose tissue, and the central nervous system, while GLP-1 receptors are found in pancreatic islets, the gastrointestinal tract, and the brain. Activation of both receptors can enhance glucose-dependent insulin secretion and reduce glucagon release. The relative contribution of each receptor to the overall pharmacological effect remains an area of ongoing investigation. Preclinical studies suggest that GIP receptor agonism may modulate appetite and energy balance, but the precise mechanisms in humans are not fully established.
In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
As a peptide, tirzepatide is handled as a lyophilised solid in research settings and as a preserved solution in finished products. Aqueous solubility is pH dependent and reaches a minimum near the isoelectric point, which lies close to pH 5.4. Stock solutions are typically prepared in neutral or slightly basic buffer to limit precipitation. The solid is hygroscopic and should be equilibrated to room temperature before opening so that condensation does not form on the powder surface.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.
== Relationship between atomic and molecular masses == Similar definitions apply to molecules. One can calculate the molecular mass of a compound by adding the atomic masses (not the standard atomic weights) of its constituent atoms. Conversely, the molar mass is usually computed from the standard atomic weights (not the atomic or nuclide masses). Thus, molecular mass and molar mass differ slightly in numerical value and represent different concepts. Molecular mass is the mass of a molecule, which is the sum of its constituent atomic masses. Molar mass is an average of the masses of the constituent molecules in a chemically pure but isotopically heterogeneous ensemble. In both cases, the multiplicity of the atoms (the number of times it occurs) must be taken into account, usually by multiplication of each unique mass by its multiplicity.
Valsartan/hydrochlorothiazide, sold under the brand name Diovan HCT among others, is a medication used to treat high blood pressure when valsartan is not sufficient. It is a combination of valsartan, an angiotensin receptor blocker with hydrochlorothiazide, a diuretic. It is taken by mouth. Common side effects include dizziness and headaches. Serious side effects may include allergic reactions, electrolyte abnormalities, and glaucoma. Use in pregnancy is not recommended. The combination was approved for medical use in the United States in 1998. It is available as a generic medication. In 2021, it was the 224th most commonly prescribed medication in the United States, with more than 1 million prescriptions.
=== Plant source === Generally, honey is classified by the floral source of the nectar from which it was made. Honeys can be from specific types of flower nectars or can be blended after collection. The pollen in honey is traceable to floral source and therefore region of origin. The rheological and melissopalynological properties of honey can be used to identify the major plant nectar source used in its production.
== Description == The house cricket is typically gray or brownish in color, growing to 16–21 millimetres (0.63–0.83 in) in length. Males and females look similar, but females will have a brown-black, needle-like ovipositor extending from the center rear, approximately the same length as the cerci, the paired appendages towards the rear-most segment of the cricket. On males, the cerci are more prominent.
Wenger appointed Vieira as club captain following Adams' retirement, and made few additions to his double-winning squad, signing defender Pascal Cygan, midfielder Gilberto Silva and utility player Kolo Touré. He was confident his side would retain the league and told the media: "Nobody will finish above us in the league. It wouldn't surprise me if we were to go unbeaten for the whole of the season". Arsenal set a new top-flight record in their opening game of the 2002–03 season; a 2–0 victory against Birmingham City was their 14th league win in succession. They continued their fine start by overhauling Nottingham Forest's top-flight record of 22 away league matches without defeat and later surpassed Manchester United's Premier League total of 29 matches unbeaten. Arsenal lost to Everton in October 2002, which began a run of four matches without a win. Though Wenger's team built an eight-point lead over challengers Manchester United by March 2003, they were overhauled in the final weeks of the season. The title race shifted in United's favour when Bolton Wanderers came from behind to draw 2–2 against Arsenal. Wenger showed moments of exasperation during the match by loosening his tie and bellowing orders from the touchline. Journalist Phil McNulty wrote that Wenger's demeanour "spoke of someone who had the title in his grasp but now saw it slipping from his fingers". Arsenal beat Southampton 1–0 to retain the FA Cup at the end of the campaign. During pre-season, Wenger arranged a meeting with the players to examine why they lost the league.

During the socialist era in Yugoslavia, the only legal political parties were the three branches of the federal League of Communists of Yugoslavia (SKJ): League of Communists of Serbia (SKS), League of Communists of Vojvodina (SKV) and League of Communists of Kosovo (SKK). The Serbian branch remained relatively stable and loyal to the federal party until the late 1980s, when it became split over what action to take in Kosovo when protests and fights broke out between ethnic Albanians and Serbs. The more traditional Communists supported President Ivan Stambolić, who advocated continued neutrality as a means to solve the dispute; while more radical and nationalist-leaning members supported Slobodan Milosević, who advocated the protection of Kosovo Serbs, who had claimed that their population was being pressured to leave Kosovo by Albanian separatists. Milosević utilized public sentiment and opposition to Kosovo Albanian separatism to rally large numbers of supporters to help him overthrow the Communist leadership in Vojvodina, Kosovo and the Socialist Republic of Montenegro in what was known as the anti-bureaucratic revolution. Afterwards, the Serbian League of Communists selected Milosević as its leader. Milosević took a hard stand on Albanian nationalism in Kosovo and pressured the Yugoslav government to give him emergency powers to deal with Kosovo Albanian separatists. Furthermore, he reduced the autonomy of the autonomous provinces of Kosovo and Vojvodina and installed politicians loyal to him to serve as their representatives.
The suffix -thiol is added to the name of the alkane. This method is nearly identical to naming an alcohol and is used by the IUPAC, e.g. CH3SH would be methanethiol. The word mercaptan replaces alcohol in the name of the equivalent alcohol compound. Example: CH3SH would be methyl mercaptan, just as CH3OH is called methyl alcohol. The term sulfhydryl- or mercapto- is used as a prefix, e.g. mercaptopurine.
=== Recreational === Amphetamine, specifically the more dopaminergic dextrorotatory enantiomer (dextroamphetamine), is also used recreationally as a euphoriant and aphrodisiac, and like other amphetamines, is used as a club drug for its energetic and euphoric high. Dextroamphetamine (d-amphetamine) is considered to have a high potential for misuse in a recreational manner since individuals typically report feeling euphoric, more alert, and more energetic after taking the drug. A notable part of the 1960s mod subculture in the UK was recreational amphetamine use, which was used to fuel all-night dances at clubs like Manchester's Twisted Wheel. Newspaper reports described dancers emerging from clubs at 5 a.m. with dilated pupils. Mods used the drug for stimulation and alertness, which they viewed as different from the intoxication caused by alcohol and other drugs. Dr. Andrew Wilson argues that for a significant minority, "amphetamines symbolised the smart, on-the-ball, cool image" and that they sought "stimulation not intoxication [...] greater awareness, not escape" and "confidence and articulacy" rather than the "drunken rowdiness of previous generations." Dextroamphetamine's dopaminergic (rewarding) properties affect the mesocorticolimbic circuit; a group of neural structures responsible for incentive salience (i.e., "wanting"; desire or craving for a reward and motivation), positive reinforcement and positively-valenced emotions, particularly ones involving pleasure. Large recreational doses of dextroamphetamine may produce symptoms of dextroamphetamine overdose.
=== α/β === α/β proteins are a class of structural domains in which the secondary structure is composed of alternating α-helices and β-strands along the backbone. The β-strands are therefore mostly parallel. Common examples include the flavodoxin fold, the TIM barrel and leucine-rich-repeat (LRR) proteins such as ribonuclease inhibitor.

== With herbal medicines == Herb-drug interactions are drug interactions that occur between herbal medicines and conventional drugs. These types of interactions may be more common than drug-drug interactions because herbal medicines often contain multiple pharmacologically active ingredients, while conventional drugs typically contain only one. Some such interactions are clinically significant, although most herbal remedies are not associated with drug interactions causing serious consequences. Most catalogued herb-drug interactions are moderate in severity. The most commonly implicated conventional drugs in herb-drug interactions are warfarin, insulin, aspirin, digoxin, and ticlopidine, due to their narrow therapeutic indices. The most commonly implicated herbs involved in such interactions are those containing St. John's Wort, magnesium, calcium, iron, or ginkgo.

March 31 2021 Orange, California office shooting – Four people are killed and two others, including the suspect, are injured in a shooting at an office building. President Biden unveils a $2 trillion infrastructure plan. Governor Greg Gianforte signs a bill that bans Sanctuary cities in the state of Montana into law. Montana becomes the 13th state to ban sanctuary cities.
=== Legal === This technique is also used for detection of illicit drugs in various samples. The most common method of drug detection has been an immunoassay. This method is much more convenient. However, convenience comes at the cost of specificity and coverage of a wide range of drugs, therefore, HPLC has been used as well as an alternative method. As HPLC is a method of determining (and possibly increasing) purity, using HPLC alone in evaluating concentrations of drugs was somewhat insufficient. Therefore, HPLC in this context is often performed in conjunction with mass spectrometry. Using liquid chromatography-mass spectrometry (LC-MS) instead of gas chromatography-mass spectrometry (GC-MS) circumvents the necessity for derivitizing with acetylating or alkylation agents, which can be a burdensome extra step. LC-MS has been used to detect a variety of agents like doping agents, drug metabolites, glucuronide conjugates, amphetamines, opioids, cocaine, BZDs, ketamine, LSD, cannabis, and pesticides. Performing HPLC in conjunction with mass spectrometry reduces the absolute need for standardizing HPLC experimental runs.
== Research == In the early 21st century, extensive research is occurring in the field of nanoparticle drug delivery systems to the brain. One of the common diseases being studied in neuroscience is Alzheimer's disease. Many studies have been done to show how nanoparticles can be used as a platform to deliver therapeutic drugs to these patients with the disease. A few Alzheimer's drugs that have been studied especially are rivastigmine, tacrine, quinoline, piperine, and curcumin. PBCA, chitosan, and PLGA nanoparticles were used as delivery systems for these drugs. Overall, the results from each drug injection with these nanoparticles showed remarkable improvements in the effects of the drug relative to non-nanoparticle delivery systems. This possibly suggests that nanoparticles could provide a promising solution to how these drugs could cross the BBB. One factor that still must be considered and accounted for is nanoparticle accumulation in the body. With long-term and frequent injections that are often required to treat chronic diseases such as Alzheimer's disease, polymeric nanoparticles could potentially build up in the body, causing undesirable effects. This area for concern would have to be further assessed to analyze these possible effects and to improve them.

It is a synthetic peptide of 39 amino acids bearing a lipid side chain. Its size and architecture place it outside the small-molecule class, and laboratories generally handle it with the precautions used for biologic-like molecules.
The fatty diacid side chain promotes strong binding to serum albumin, which slows clearance and yields a half-life of roughly five days. That profile supports once-weekly administration in clinical use.
In vitro assays detect activity at both the GIP and GLP-1 receptors, but the activity ratio depends on the assay system and the signalling pathway measured. The relative contribution of each receptor to clinical effects is still being characterised.
It is normally kept frozen, desiccated, and away from light, with brief warming to room temperature before opening to limit condensation. Repeated freeze-thaw cycles are avoided because they stress the peptide. Once in solution, the material is held cold and used promptly.
