The short version of deamidation fits in a sentence. The long version — which is the one that helps — is below.

Regulatory and quality discussions place the peptide within established guidance for synthetic peptides and biologics. Forced degradation studies, in which samples are exposed to heat, acid, base, peroxide, and light, identify likely degradation products and validate the selectivity of analytical methods. Reference standards allow comparison across laboratories and production batches. Purity specifications reported in the literature usually combine chromatographic purity with mass confirmation. Which impurity thresholds are meaningful for long-term behavior is still debated, and no single universal specification has been adopted across all jurisdictions.
Routine characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity assessment, usually with ultraviolet detection near 214 nanometers. Intact mass measurement by liquid chromatography coupled to mass spectrometry confirms molecular identity against a theoretical value. Sequence-level confirmation uses enzymatic digestion followed by tandem mass spectrometry, an approach known as peptide mapping. Amino acid analysis gives an independent check on composition. Circular dichroism spectra are used to estimate helical content in aqueous buffer.
Stability depends strongly on physical form. The dry powder is generally regarded as stable for extended periods when held at or below minus twenty degrees Celsius in a sealed, desiccated container. In solution, degradation pathways include deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation. Reaction rates for these pathways rise with temperature. Repeated freezing and thawing of solutions promotes aggregation, and light exposure can accelerate some oxidative changes. Buffer composition and pH influence which pathway dominates at a given temperature.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.

Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Research and analytical settings increasingly require documentation of peptide origin and chain of custody. Certificate of analysis documents typically report purity by chromatographic area, mass confirmation, appearance, and residual solvent or counterion content. Independent verification by an accredited laboratory is common when a material will be used in a regulated study. Open questions remain about how well compendial methods transfer between laboratories, and about which impurity thresholds are meaningful for materials not intended for clinical use.
Peptide-based pharmaceutical products such as tirzepatide require controlled temperature management to preserve structural integrity. Manufacturer labeling generally specifies refrigeration at 2 to 8 degrees Celsius before first use, with protection from light and freezing. Exposure to repeated temperature cycling can promote aggregation or deamidation, which alters the analytical profile even when the visible solution appears unchanged. Once a product is in use, the permitted storage window and temperature range are defined by the specific labeled presentation rather than by general peptide rules.
Identity and purity assessment of tirzepatide relies primarily on reversed-phase high-performance liquid chromatography coupled with ultraviolet detection. Mass spectrometry, often in electrospray ionization mode, confirms the molecular mass and detects sequence-related impurities. Peptide mapping after enzymatic digestion provides residue-level confirmation of the backbone. Each method addresses a different question: chromatography for purity and related substances, mass measurement for identity, and mapping for sequence fidelity. No single technique covers all three.
==== Abolition of the state ==== Socialist Studies accuses the SPGB of coming under the influence of anarchist ideas and not sufficiently emphasising the parliamentary aspect of the socialist revolution. Socialist Studies claims that the SPGB's position is that the state would be abolished immediately upon the overthrow of class society whereas Socialist Studies's position is that the state would "gradually wither away" instead. Referring to the SPGB's 1984 conference resolution (since rescinded) that "Socialism will entail the immediate abolition of and not the gradual decline of the State", Socialist Studies writes: If you are in favour of the immediate abolition of the State when Socialism is established then you are in favour of its immediate abolition here and now, which is of course the anarchist position. [...] Unless Socialists use the power of the machinery of government to dispossess the capitalist class they will be unable to establish Socialism. We find it inconceivable that intelligent Socialists of a future generation would even consider such an act of monumental stupidity.
The Congress of Vienna instituted a territorial adjustment between Hanover and Prussia to form more contiguous borders. Hanover increased its area substantially, gaining the Prince-Bishopric of Hildesheim, East Frisia, the Prince-Bishopric of Osnabrück, the Lower County of Lingen and the northern part of the Prince-Bishopric of Münster. Hanover also annexed territories that had previously been ruled in personal union by its Elector, such as the Duchies of Bremen-Verden and the County of Bentheim. It lost those parts of Saxe-Lauenburg to the northeast of the Elbe, which was assigned in personal union to Denmark, except the Amt Neuhaus. Further small exclaves in the east were lost. Hanover comprised a number of territories, which had been Imperial Estates within the Holy Roman Empire. Their respective governments, now called provincial governments, were organised according to partially very old traditions, including different levels of estate participation in rule. In 1823, the kingdom was reorganised into high-bailiwicks (German: Landdrosteien, singular: Landdrostei), each led by a high-bailiff (German: Landdrost) according to unitary standards, doing away with the inherited provincial peculiarities. The high-bailiwicks were subdivided into bailiwicks (German: Ämter, singular Amt), presided by a bailiff (Amtmann, plural Amtleute). The high-bailiwicks, named after their capitals, were the following:
Both ROS-elevating and ROS-eliminating strategies have been developed with the former being predominantly used. Cancer cells with elevated ROS levels depend heavily on the antioxidant defense system. ROS-elevating drugs further increase cellular ROS stress level, either by direct ROS-generation (e.g. motexafin gadolinium, elesclomol) or by agents that abrogate the inherent antioxidant system such as SOD inhibitor (e.g. ATN-224, 2-methoxyestradiol) and GSH inhibitor (e.g. PEITC, buthionine sulfoximine (BSO)). The result is an overall increase in endogenous ROS, which when above a cellular tolerability threshold, may induce cell death. On the other hand, normal cells appear to have, under lower basal stress and reserve, a higher capacity to cope with additional ROS-generating insults than cancer cells do. Therefore, the elevation of ROS in all cells can be used to achieve the selective killing of cancer cells. James Watson and others have proposed that lack of intracellular ROS due to a lack of physical exercise may contribute to the malignant progression of cancer, because spikes of ROS are needed to correctly fold proteins in the endoplasmic reticulum and low ROS levels may thus aspecifically hamper the formation of tumor suppressor proteins. Since physical exercise induces temporary spikes of ROS, this may explain why physical exercise is beneficial for cancer patient prognosis.
Pseudomonas syringae pv. tabaci, the causal agent of the wildfire of tobacco P. syringae pv. coronafaciens P. syringae pv. garcae P. syringae BR2, which causes a disease of bean (Phaseolus vulgaris) similar to tobacco wildfire. This organism is closely related to P. syringae pv. tabaci but cannot be classified in the pathovar tabaci because it is not pathogenic on tobacco. Tabtoxin is a dipeptide precursor to the biologically active form of TBL, differing by having an extra threonine attached by a peptide bond to the C terminus. Tabtoxin is required by BR2(R) for both chlorosis and lesion formation on bean. All mutations that affected tabtoxin production, whether spontaneous deletion or transposon-induced, also affected lesion formation, and in all cases, restoration of tabtoxin production also restored pathogenic symptoms. Other factors may be required for BR2 to be pathogenic on bean, but apparently these are in addition to tabtoxin production. This toxin is a irreversible inhibitor of glutamine synthetase.
In 2005, Serpin A12 was discovered to be found in rats, mice, and humans. One study confirmed that those with insulin resistance had higher levels of Serpin A12 than others. Thus, those humans (or animals) who were diabetic had more Serpin A12 than those who were not. The explanation for this lies in the fact that type 2 diabetes is related to inflammation processes, hence coinciding with the anti-inflammatory effect of Serpin A12.

== Diagnosis == A presumptive clinical diagnosis of FPLV can be made for kittens with appropriate signalment, history, clinical findings and the history of no prior vaccination. The clinical diagnosis is usually supported by documenting parvovirus antigen in feces by ELISA (enzyme-linked immunosorbent assay) and PCR (polymerase chain reaction) assays. The availability of validated assays varies by country but is becoming more common. PCR assays are so sensitive that FPV DNA can be amplified from feces of cats vaccinated with modified live strains of the virus. Attenuated parvoviruses in MLV vaccines replicate in the blood and intestine, and post-vaccinal fecal shedding of FPV has been demonstrated, which can result in recent vaccinations giving false positive results on diagnostic tests. At least one of the ELISA antigen tests for dogs (SNAP®Parvo; IDEXX Laboratories) detects FPV in feline feces and has a cut point for a positive test result that excludes most vaccinated cats. Thus, this ELISA is superior to PCR for screening cats for FPV infection and can also be performed in the veterinary clinic. (These are only approved and licensed for detecting canine parvovirus, but it is generally known that they also detect FPL viral antigen in feline feces. These tests are used extra-label because they allow rapid, inexpensive, in-house detection of the virus.) Positive fecal SNAP test results, including weak positives, are highly likely to be true positives in clinically affected animals.
==== Unanimous selection for Most Valuable Player Award (1994) ==== The most productive season in Bagwell's professional career was the strike-shortened 1994 season which set several franchise records. On Opening Day, April 4, against the Montreal Expos, Bagwell hit the game-tying home run while going 3-for-6 as the Astros won in a 12-inning walk-off. For the month of April, he batted .360 with six home runs and added another six home runs in May. In June, he hit 13 home runs, establishing an Astros' franchise record for one month, while batting .394, 11 doubles, .455 OBP, .899 SLG, and 1.354 OPS. On June 24, in a 16–4 rout of the Los Angeles Dodgers at the Astrodome, he hit three home runs, including two in the same inning, becoming the first Astro since Glenn Davis in 1990 to do so. Bagwell was the NL Player of the Week for consecutive weeks on June 19 and June 26 and the NL Player of the Month for June, his second career monthly award. Selected to his first All-Star Game as a reserve, Bagwell had tied Kirby Puckett for the major league RBI lead at 81 going into the All-Star break, and batted .348 with 27 home runs and 74 runs scored. In the All-Star Game, Bagwell entered as a pinch hitter for starting pitcher Greg Maddux, singling off David Cone, and wound up collecting two hits in four at bats. The first player in the majors to reach 100 RBI on July 27, Bagwell homered off José Rijo in a 6–5 win over Cincinnati to give him 101 RBI in his first 101 games. In July, he tied a club record for RBI in any month with 29, which José Cruz and Jimmy Wynn shared.
=== Cluster headaches === Preventive therapy with verapamil is believed to work because it has an effect on the circadian rhythm and on CGRPs, as CGRP-release is controlled by voltage-gated calcium channels.
=== Athlete biological passport === The athlete biological passport is a program that tracks the location of an athlete to combat doping in sports. This means that the athlete can be monitored and drug tested wherever they are and this data can be compared to the history of their doping test results. There is an ongoing discussion about how this measure can be seen as a violation of an individual's privacy.
==== Economic Development and trade ==== Minister Smith tabled Bill 47: Making Ontario Open for Business Act, 2018, which was passed on November 21, 2018. According to the Toronto Sun, Bill 47 strips "part-time workers of two paid sick days a year and prevent[s] a rise in the minimum wage to $15 an hour on January 1, 2019." NDP critic said that this "will incent employers to turn full-time positions into cheaper part-time work". The Ontario government abruptly cut all its annual provincial funding to the Ontario Institute for Regenerative Medicine (OIRM)—$5 million—in May 2019. Minister Smith, said that the "private sector will step up and fund stem-cell research." Scientists told CBC that the private sector will only invest in the stem-cell field when "their studies reach a late phase"; until then, "government funding is crucial." OIRM scientists who are "working on treatment of premature babies" said the cuts were "extremely short-sighted and uninformed". In June 2019, Vic Fedeli was appointed as Minister of Economic Development, Job Creation and Trade. Prabmeet Sarkaria is Associate Minister of Small Business and Red Tape Reduction in the economic development ministry. Michael Parsa and Donna Skelly were appointed as Parliamentary Assistants to the Minister of Economic Development, Job Creation, and Trade (Trade) in June 2018.

Madera Community Hospital is a not-for-profit community health resource and is not associated with any other hospital or health system. The hospital is locally governed by a board of trustees which provides governance and oversight. The board is composed of over a dozen community and business leaders. Madera Community Hospital was founded in 1971 and is located at 1250 E. Almond Avenue in Madera, California. The hospital contained 106 acute care beds, a 16-bed Emergency Department, and a 10-bed Intensive Care Unit. Madera Community Hospital also operates two rural health care clinics (Mendota and Chowchilla) and a home health agency (Family Health Services in Madera). Madera Community Hospital is fully accredited by the American Osteopathic Association's Healthcare Facilities Accreditation Program (HFAP), by the Clinical Laboratory Improvement Act, 1988 (CLIA) program, and is a member of the Hospital Council of Northern and Central California and the California Healthcare Association. The COVID-19 pandemic overwhelmed San Joaquin Valley hospitals. Already financially stressed, increasing costs for medical equipment and salaries for traveling nurses to address the surges were too much for the hospital to absorb. On January 3, 2023, Madera Community Hospital shuttered and filed for bankruptcy. American Advanced Management is currently working to reopen the facility.
=== Controversy === The British critical psychiatrist Joanna Moncrieff has critiqued the use and study of esketamine and ketamine as well as related drugs like psychedelics for treatment of psychiatric disorders, highlighting concerns including excessive hype around these drugs, questionable biologically based theories of benefit, blurred lines between medical and recreational use, flawed clinical trial findings, financial conflicts of interest, strong expectancy effects and large placebo responses, small and short-term benefits over placebo, and their potential for difficult experiences and adverse effects, among others.
Over the November 2016 Thanksgiving holiday weekend, President-Elect Donald Trump tweeted that he was in talks with Carrier Management to keep the factory in Indiana and not move to Mexico. On November 30, 2016, Carrier announced that it had negotiated an agreement with President-elect Trump and Vice President-elect Mike Pence to continue manufacturing gas furnaces in Indianapolis, in addition to retaining engineering and headquarters staff, preserving more than 1,000 jobs in Indianapolis. The agreement included a state incentive package of about $7 million over 10 years. The number of jobs saved was later revised down to 800. In May 2017, as part of their previously announced plan, Carrier told the state of Indiana that it will cut 632 jobs from its Indianapolis factory. Layoffs began at the end of July 2017, with each worker receiving one week's salary for each year of employment, education and job training, plus 6 months of health insurance as part of the severance package. The New York Times reported on August 10, 2018, that Carrier's Indianapolis furnace plant was plagued by low morale and absenteeism because "employees share a looming sense that a factory shutdown is inevitable — that Carrier has merely postponed the closing until a more politically opportune moment." On November 26, 2018, United Technologies announced that it would spin off UTC Climate, Controls & Security as an independent company known as Carrier Global Corporation. In April 2020, United Technologies announced that the separation and spin-off of Carrier had been completed.
=== Mental illness referred to in children's television === A 2000 study of New Zealand Children's television shows showed that a mental illness reference appeared in 59 out of 128 episodes chosen. In those 59 episodes, there were 159 references to mental illness, frequently of character descriptions. The terms "mad," "crazy," and "losing your mind" were above the three most common references. Character appearances consisted of disfigured facial features (teeth, noses, etc.) and disfigured extremities. Mental illness has also been portrayed in Walt Disney animated films. A study examined the portrayal of mental illness in Disney films and found that 85% of these films made reference to mental illness, and 21% of the characters were referred to as mentally ill. On average, 4.6 references to mental illness were made in these films, with the most commonly used terms being "mad," "crazy," or "nutty." The study highlighted how Maurice, from Beauty and the Beast, was often depicted as mentally ill, referred to as a lunatic, and was even attempted to be sent to an asylum. Another study examined 40 children's programs on Netflix, analyzing 339 episodes for references to mental illness. The study found that 23 of these programs had at least one reference to mental illness, with 58 episodes featuring such references. It also noted differences in how male and female characters with mental illness were portrayed. Male characters were often depicted as violent or aggressive, while female characters were shown as "crazy" or obsessive.
Prior to 1999, the Engineer designations were 12 series and 83 series. In 1999, CMF 83 changed to CMF 51. In 2004, CMF 51 changed to CMF 21. In 2004, the engineer designation changed from 12 to 21. In 2009, the engineer designation was changed again, from CMF 21 to CMF 12. In 2013, the engineer officer designations 12B (Combat Engineer) and 12D (Facilities/Contract Construction Management Engineer (FCCME)) were consolidated into 12A.

Peptide mapping with tandem mass spectrometry is the standard approach. The peptide is digested with an enzyme such as trypsin, and the resulting fragments are matched against the expected sequence.
Higher pH and elevated temperature both increase deamidation rates. Holding solutions at low temperature and near-neutral to slightly acidic pH reduces the extent of the reaction.
Removing water slows hydrolysis and aggregation. The dry powder tolerates longer storage intervals than a solution kept at the same temperature.
Reversed-phase liquid chromatography with ultraviolet detection is the usual approach, frequently combined with mass spectrometry for identity. Purity is reported as the area percentage of the main peak. Related impurities eluting near the main peak are usually summed and reported separately.
